Speaker Talk title Extracellular Vesicles in Alzheimer’s Disease: From Plaque Pathology to Neuronal Network Function. Abstract Extracellular vesicles (EVs) are emerging as important mediators in the pathophysiology of sporadic Alzheimer’s disease (AD), carrying diverse bioactive cargo including RNA and proteins. In our work, we investigate the role of EVs in amyloid-β (Aβ) aggregation and plaque association by combining analyses of human post-mortem brain tissue, multiple amyloid mouse models, and cellular systems with biochemical and structural biology approaches. Our findings indicate that EVs carrying the cellular prion protein (PrPC) promote Aβ aggregation and contribute to plaque association in Alzheimer’s disease. In addition, we characterized disease-specific cargo signatures in human AD brain-derived EVs and examine their functional effects on neuronal network formation using human iPSC-derived cortical neurons.
